Description
Specificity:
Anti-Human Interluekin-2(IL-2) (Clone L032) recognizes Human Interluekin-2. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.
Background:
Interleukin-2 (IL-2) is a potent T-cell mitogenic cytokine1 that functions as a primary regulator of T cell homeostasis and has been considered a prime candidate immunotherapeutics, both for increasing T cell levels/function and for augmenting vaccine-elicited viral-specific T cell responses.2 IL-2 plays a key role in setting the balance between immunity and tolerance. This cytokine has a dual role as the regulator of the two main phases of the immune response (proliferative and suppressive).3 IL-2 also stimulates growth and differentiation of B cells, NK cells, LAK cells, monocytes, and oligodendrocytes.

Formulation
This purified antibody is formulated with no carrier proteins and lyophilzed from phosphate buffered saline (PBS).
Reconstitution
This monoclonal antibody should be aseptically reconstituted in sterile deionized water to a concentration of 1.0 mg/ml.
Storage and Stability
This lyophilized monoclonal antibody should be stored desiccated at -20oC. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20oC in a manual defrost freezer. Avoid Repeated Freeze Thaw Cycles.

Indirect ELISA, IHC (NBF/Par.), FC, N
Indirect ELISA: This antibody can be used in an indirect detection ELISA at 0.5 - 1.0 µg/ml with a suitable second step reagent such as Goat Anti-Mouse IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: M114). The detection sensitivity of this indirect ELISA for Human IL-2 is approximately 5 ng/well.
IHC (NBF/Par.): This antibody should give satisfactory staining results when used at a concentration of 8-25 µg/ml. The recommended secondary antibody for IHC is Goat Anti-Mouse IgG (H&L)-Horseradish Peroxidase (Leinco Prod. No.: M114). For chromogenic detection with high signal and low background use 3, 3-Diaminobenzidine Tetrahydrochloride (DAB) Substrate Kit (Leinco Prod. No.: D100) or Optional Metal Enhanced DAB Substrate Kit (Leinco Prod. No.: K107).
Flow Cytometry: It is recommended to use the indirect method for signal enhancement when enumerating cells expressing IL-2. A suggested method would be to stain cells expressing IL-2 with 0.25 µg per 1-5 x 105 cells in a total staining volume of µl followed by Goat Anti-Mouse IgG (H&L)-R-phycoerythrin (Leinco Prod. No.: M1189).
Neutralization: This antibody is useful for neutralization of Human IL-2 bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Human IL-2 (at 2 ng/ml) is 0.015 - 0.03 µg/ml.
